The investigation compares heart rate under a defined caffeine treatment with an appropriate water or vehicle control. Count beats over a stated interval using the approved observation method, then convert to beats per minute. An actual count and timing rule is more reproducible than describing a heart as “faster”. High magnification or strong illumination can warm the preparation, so observation conditions are part of the design.

Viewing focus: Design a controlled invertebrate heart-rate comparison
After watching, attempt these questions before opening the answers. You can also use the written lesson below.
After watching 1: Why control illumination during observation?
It can alter temperature or the animal’s conditions. The observation method can itself change the response.
After watching 2: What distinguishes a biological replicate here?
An appropriately independent animal. Repeated counts and independent individuals represent different variation.
What you'll be able to do
The investigation compares heart rate under a defined caffeine treatment with an appropriate water or vehicle control. Count beats over a stated interval using the approved observation method, then convert to beats per minute. An actual count and timing rule is more reproducible than describing a heart as “faster”. High magnification or strong illumination can warm the preparation, so observation conditions are part of the design.
Control temperature, solution volume, exposure time, handling and other relevant conditions. Select the teacher-approved treatment range and acclimation procedure. Do not infer a human dose or a human cardiovascular effect from a classroom invertebrate response. The evidence concerns the organism and conditions actually studied.
Repeated counts from one animal can assess counting variation but share that individual’s physiology and treatment history. Independent animals are needed to estimate variation among animals. If the same animal receives more than one condition, use a justified repeated-measures design with appropriate order, recovery and analysis rather than treating every count as a new organism.
Randomise or counterbalance treatment order where suitable and use blinded counting from an appropriate recording when feasible. A control observed only at the beginning cannot separate a treatment effect from a later temperature or time drift. Record raw counts and intervals, not only rounded rates or selected best-looking clips.
Invertebrate use still raises ethical questions. Minimise exposure, disturbance and unnecessary repetition, use only the approved handling procedure, and follow the centre’s decisions about recovery and care. The small body size and high heartbeat rate can make counting uncertain, so an unsuitable observation is a reason to improve measurement rather than expose an animal repeatedly without purpose.
Compare mean rates and an appropriate measure of variation using the actual independent sample size. A difference in a small dataset does not automatically prove a universal caffeine effect. Explain plausible biological responses while considering individual differences, observation bias and residual confounding. Report the limits of the conclusion alongside the numerical comparison.
Attempt these problems on paper before opening the worked reasoning. Explain your steps as well as giving a final answer.

Key takeaways
Question 1: Three counts are taken from each of four animals in one treatment. How many independent animals were observed?
Four independent animals, not twelve independent animals.
